The world number one destroyed American Chris White 6-0 in just 11 minutes as he averaged 104.86 and hit six of his eight attempts at a double to breeze into the quarter-finals.
And van Gerwen finished the match off in showboating style by nailing back-to-back bullseyes and double 16 to take out 132.
Gary Anderson took just a couple of minutes longer to whitewash Jayson Barlow 6-0 with an average of 102.48 in the final match of the night but the North American qualifiers had earlier given a much better account of themselves in the opening four games of this two-day World Series of Darts tournament, which concludes on Saturday.
Roared on by a raucus crowd at the Tropicana Hotel Casino Resort, young Canadian Dawson Murschell produced a huge shock as he knocked out world number six Wade.
The Machine looked like he would avoid the scare when presented with five darts at his favourite double - tops - in the deciding leg but he missed them all as well as one at double 10 and Murschell took full advantage to spark wild celebrations.
Germany's number one Max Hopp was the other invited player to be knocked out as he suffered a 6-3 defeat against the impressive Dave Richardson, who was runner-up in the North American Championship 24 hours earlier.
The man who beat Richardson, Willard Bruguier, also gave a good account of himself as he forced Gerwyn Price to dig deep for a 6-4 win.
Raymond van Barneveld was also given a tough workout by David Cameron but won three legs in succession to win through 6-3 while it was the same story for Peter Wright in his clash with Canada's Shawn Brenneman.
Daryl Gurney made a solid start to his World Series of Darts debut with a 6-2 victory over Dennis Sayre.
Saturday will see the quarter-finals, semi-finals and final contested, with all games the first to eight legs.
